56 (3). Another recent report concerned with the incidence of respiratory cancer in nickel workers refers to a total cancer mortality excess of over 3 times that in the population as a whole during the 9-year period 19@9—38, this excess occurring entirely in the two sites referred to above (11). The latter report also points out that in the Clydach Works, “Nodeaths from nasal cancer have been recorded in workers engaged after 19@4, at which date increased precautions were taken against dust.― The drying and powdering of cop per sulfate and the arsenic content of the sulfuric acid used before 19@1 are suggested as being

Role of metals in carcinogenesis. Experimental studies in whole animal bioassay.

Animal models have been used for a number of purposes in metal carcinogenesis studies: (a) to detect carcinogenic activity; (b) to estimate carcinogenic risk; and (c) to investigate mechanisms of metal carcinogenesis. The design of animal tests will be determined by the purpose of the study. In this section we shall distinguish between tests in animals designed to identify carcinogenic activity...

Peptide models in the study of the mechanism of carcinogenesis by heavy metals*

The molecular mechanisms of carcinogenesis involving heavy metal ions are not yet fully understood. Histones surrounding DNA are believed to be primary targets for metal ion binding, and such interactions may play a direct or indirect role in metal-induced toxicity carcinogenesis. This paper reviews our results of approximately the last 10 years in this area, starting from small peptide fragmen...

Active oxygen species in DNA damage induced by carcinogenic metal compounds.

Some carcinogenic metal compounds [chromate(VI), Fe(III) nitrilotriacetate, cobalt(II), and nickel(II)[ induced formation of various oxygen radical species in the presence of hydrogen peroxide. These oxygen radicals were suggested to give different kinds of site-specific DNA damage; 8-hydroxyl-2'-deoxyguanosine formation is included in the DNA damage. Using pulsed-field gel electrophoresis, nic...

Aneuploidy as an early mechanistic event in metal carcinogenesis.

Aneuploidy has recently been proposed as an initiating event for carcinogenesis. There is significant evidence that carcinogenic metals induce aneuploidy. Here we review the mechanisms for how carcinogenic metals may induce aneuploidy and the evidence that carcinogenic metals induce an aneugenic effect which can destabilize the genome leading to genomic instability and cancer.

Epigenetics in metal carcinogenesis: nickel, arsenic, chromium and cadmium.

Although carcinogenic metals have been known to disrupt a wide range of cellular processes the precise mechanism by which these exert their carcinogenic effects is not known. Over the last decade or two, studies in the field of metal carcinogenesis suggest that epigenetic mechanisms may play a role in metal-induced carcinogenesis. In this review we summarize the evidence demonstrating that expo...

Epidemiological and experimental aspects of metal carcinogenesis: physicochemical properties, kinetics, and the active species.

The carcinogenic properties of selected metals and their compounds are reviewed to provide a useful reference for existing knowledge on relationships between physical and chemical forms, kinetics and carcinogenic potential and between epidemiology, bioassays, and short-term tests. Extensive consideration is given to arsenic, beryllium, cadmium, chromium, lead, and nickel. Other metals such as a...
